A Clinical Case of Severe Anemia in a Sheep Coinfected with Mycoplasma ovis and 'Candidatus Mycoplasma haemovis' in Hokkaido, Japan OAK

A 2-year-old East Friesian sheep imported from Australia exhibited severe anemia after contagious pustular dermatitis in Hokkaido, Japan. Hemoplasma infection was confirmed in blood smears. Both Mycoplasma ovis and ‘Candidatus Mycoplasma haemovis’ were detected by PCR and sequence analyses. In the epidemiological analysis, dual pathogens were detected in 6 of 12 (50.0%) sheep imported from Australia with the infected ewe at the same time, 1 of 5 (20.0%) sheep introduced from a domestic farm in Hokkaido, and in 1 of 16 (6.3%) sheep from an epidemiologically unrelated ranch. It is the first clinical case of sheep to confirm coinfection of these pathogens in Japan.

Cerebellar Cortical Degeneration in a Japanese Black Bull OAK

黒毛和種牛が出生翌日より横臥,後弓反張を呈した.脳挫傷を疑い,抗生物質,ビタミンB1製剤,消炎鎮痛剤により加療され,いったんは改善したが同様の症状が間欠的に認められた.歩様異常は継続して認められたが,一般状態は良好で8カ月齢まで飼養された.しかし,その後開脚姿勢,測定過大を伴う歩様異常,威嚇瞬き反応の低下が進行した.病理組織学的検索により小脳にプルキンエ細胞の顕著な変性と脱落及び顆粒細胞の減数を認め,小脳皮質変性症と診断された.

Detection of Anaplasma bovis DNA in the Peripheral Blood of Domestic Dogs in Japan OAK

The prevalence of Ehrlichia and Anaplasma in 1,427 dogs from 32 Japanese prefectures was evaluated by PCR and DNA nucleotide sequencing. PCR screening demonstrated that 18 dogs (1.3%) were positive for Anaplasmataceae. Sequence analysis revealed that 14 of the amplicons were most closely related to Wolbachia spp., symbionts of Dirofilaria immitis, whereas three were identified as Anaplasma bovis. The remaining amplicon could not be sequenced. Bovine dilated cardiomyopathy (DCM) is an autosomal recessive genetic disorder causing congestive heart failure and subsequent death. Recently, a nonsense mutation c.343C>T in the bovine optic atrophy 3 (OPA3) gene had been reported to cause the DCM in Holstein cattle in Switzerland. However, the mutation has not been confirmed in bovine DCM outside Switzerland. Nine Holstein Friesian cows that were macroscopically and histologically diagnosed with or suspected of DCM and 12 control cows kept in Japan were tested for the mutation. Anaplasma phagocytophilum causes human granulocytic anaplasmosis (HGA) and tick-borne fever in ruminants. A closely related and potentially novel Anaplasma sp. in Japan was recently characterized. The present study evaluated the effect of hemoplasmosis on cattle productivity. Prevalence of bovine hemoplasma was examined by polymerase chain reaction (PCR) using whole blood samples collected from 93 breeding cows and their 71 calves in Hokkaido, Japan. Monthly milk production records and other clinical data were compared between Mycoplasma wenyonii (Mw)-infected, “Candidatus Mycoplasma haemobos” (CMh)-infected, co-infected and PCR-negative groups. Anaplasma marginale has been detected in the Philippines only by peripheral blood smear examination and serological methods. This study generally aimed to molecularly detect and characterize A. marginale in cattle and ticks in Cebu, Philippines. A total of 12 bovine blood samples and 60 Rhipicephalus (Boophilus) microplus ticks were collected on the Cebu Island in 2011. 16S rRNA-based screening-PCR and DNA sequencing revealed 8 cattle (66.7%) and 8 ticks (13.3%) to be positive for A. marginale, and 1 tick (1.7%) to be positive for A. centrale. Theileria orientalis is a benign protozoan species that is widely distributed in Japan, yet sometimes causes serious economic losses in the livestock industry. In this study, we conducted a molecular survey based on genes encoding the major piroplasm surface protein (MPSP) and p23 for T. orientalis detected in cattle grazing in southern areas of Japan, consisting of 2 farms in Kumamoto prefecture (Aso and Kuma districts) and 3 farms in Okinawa prefecture (Ishigaki, Iriomote, and Yonaguni Islands). Glomus tumor of the liver in a cow OAK

An 11-year-old Holstein-Friesian cow exhibited anorexia and jaundice. A large mass was found in the liver during necropsy. Macroscopically, the mass was composed of dark red multilobular tissue and a centrally located abscess, which was connected to the hepatic duct. Histologically, the mass consisted of proliferation of small neoplastic cells and was demarcated from the hepatic parenchyma by a thick region of granulation tissue. 10歳の雄の雑種犬が,3週間前から持続する発熱及び元気消失を主訴に帯広畜産大学動物医療センターを受診した.心雑音及び心エコー検査において僧帽弁の疣贅物が確認され,抗生物質等により加療したが,第7病日より心不全の悪化を認め,第10病日に肺水腫により斃死した.病理解剖では,軽度の心拡大及び僧帽弁における多中心性疣贅物が認められた.血液培養検査では,Staphylococcus haemolyticusが検出されたが,疣贅物を用いたPCR検査ではStreptococcus canisと100%一致する塩基配列が得られた.血液培養分離菌はコンタミネーションであった可能性が高く,本症例の心内膜炎はStreptococcus canisによるものと考えられた.血液培養結果と実際の心内膜炎起因菌が異なる場合があることを考慮する必要がある.

Theileria orientalis is a benign bovine protozoan parasite that occasionally causes serious economic loss in the livestock industry. We report the findings of a molecular epidemiological survey of T. orientalis in 94 Vietnamese yellow cattle, 43 water buffaloes, 21 sheep, 21 goats and 85 blood-sucking ticks of cattle in the Thua Thien Hue province of Vietnam. The major piroplasm surface protein (MPSP) gene of T. orientalis was detected using polymerase chain reaction from 13 cattle (13.8%), 11 water buffaloes (25.6%), 1 sheep (4.8%) and 9 ticks (10.6%). Phylogenetic analysis using MPSP gene sequences showed the presence of seven genotypes, four previously categorized genotypes (Types 1, 3, 5 and 7) and three new genotypes (Types N-1, N-2 and N-3).

Molecular Survey of Rickettsial Agents in Feral Raccoons (Procyon lotor) in Hokkaido, Japan OAK

Rickettsial infection in feral raccoons (Procyon lotor) in Hokkaido, Japan was analyzed by molecular methods. Genus-specific nested polymerase chain reaction (PCR) analysis based on the Rickettsia citrate synthase (gltA) gene showed that 13 of 699 raccoons (1.9%) examined were positive for Rickettsia. Twelve of the 13 partial gltA sequence amplicons were successfully analyzed. The nucleotide sequence of one amplicon was identical to both Rickettsia heilongjiangensis and R. japonica, one was identical to R. felis, and the rest to R. helvetica. This is the first report on the detection of rickettsial agents in peripheral blood of raccoons.

Degeneration of the optic pathway has been reported in various animal species including cattle. We experienced a case of bilateral optic tract degeneration characterized by severe gliosis in a Japanese black cattle without any obvious visual defects. To evaluate the significance, pathological nature and pathogenesis of the lesions, we examined the optic pathway in 60 cattle (41 Japanese black, 13 Holstein and 6 crossbreed) with or without ocular abnormalities. None of these animals had optic canal stenosis. Perforating abomasal ulcer caused by yolk sac tumor in a Holstein calf OAK

A 2-month-old male Holstein calf showed clinical signs of abdominal bloating, melena, and pain and was suspected of having a perforating abomasal ulcer. Necropsy revealed a large mass located preferentially around the abomasum and a large perforating abomasal ulcer on the pyloric antrum. Milky white masses of various sizes were also found in the abdominal cavity that consisted of agglutinated nodules ranging in size from a pinhead to a golf ball and were distributed on the surfaces of the liver, reticulum, omasum, abomasum, and diaphragm. Prevalence and Molecular Analyses of Hemotrophic Mycoplasma spp. (Hemoplasmas) Detected in Sika Deer (Cervus nippon yesoensis) in Japan OAK

Hemotropic mycoplasmas (hemoplasmas) are cell-wall deficient, epierythrocytic bacteria that cause infectious anemia in several mammalian species. The prevalence of hemoplasma species was examined by screening and species-specific PCR using blood samples collected from 51 sika deer in Hokkaido, Japan. Molecular analyses were performed for the 16S rRNA, 23S rRNA and RNase P RNA (rnpB) gene sequences. A total of 23/51 (45%) deer DNA samples were positive for hemoplasmas in the screening PCR. Prevalence and Risk Factor Analysis of Bovine Hemoplasma Infection by Direct PCR in Eastern Hokkaido, Japan OAK

The prevalence and risk factors of 2 bovine hemoplasma species—Mycoplasma wenyonii (Mw) and “Candidatus Mycoplasma haemobos” (CMh)—were examined by direct PCR using whole blood samples collected from 343 cattle in Eastern Hokkaido, Japan. The sensitivity of the direct PCR assay was 10-fold higher than the standard PCR assay, detecting 5 copies of bovine hemoplasma DNA per reaction. An epidemiological survey of sampling location, living conditions, age and bovine leukemia virus antibody was performed to assess infection risk factors. Anaplasma marginale is an etiologic agent of bovine anaplasmosis. This study aimed to molecularly detect and characterize A. marginale that is prevalent in Mongolian cattle populations. A highly specific and sensitive nested PCR (nPCR) method based on the Msp5 gene was developed to detect A. marginale (Msp5 nPCR). The method detected A. marginale from the positive DNA samples obtained from different countries, while no amplicons were observed from DNA samples of several other bovine blood pathogens tested. A total of 87 Thoroughbred horses and 10 ixodid ticks from a ranch in Hidaka district, Hokkaido were tested for tick-borne diseases. Using the indirect fluorescent antibody (IFA) method, 3.4, 92.0 and 97.7% of the horses showed antibody titers of ≥80 against Anaplasma phagocytophilum, Rickettsia helvetica, and Borrelia garinii, respectively. This is the first report of infection with the 3 pathogens in horses in Japan. Using PCR, DNAs from the peripheral blood of all horses were found negative with any Anaplasma, Rickettsia and Borrelia spp., while those from Haemaphysalis megaspinosa ticks were found positive for Anaplasma sp. closely related to A. phagocytophilum in Japan, and A. bovis. B. japonica was also detected in an H. flava tick for the first time.
